Changes in N2 fixation in Stylosanthes scabra derived from tissue culture Braz. J. Genet.

Plants were regenerated from leaf-derived callus culture of Stylosanthes scabra, a polyploid legume tolerant to drought and adapted to acid soils. A total of 168 regenerants were planted out in Leonard jars in a complete randomized design. Nitrogen fixation and vegetative growth were indirectly evaluated by shoot dry weight, root dry weight, shoot N content and acetylene reduction activity. The results showed higher variation in the regenerants than in controls not submitted to tissue culture. Significant differences were found for all nitrogen fixation related-traits

Identification of a splicing coactivator gene that affects the production of ochratoxin a in Aspergillus carbonarius BABT

Ochratoxin A is a mycotoxin produced by some fungi species. Among them, Aspergillus carbonarius is considered a powerful producer. Genes involved in the ochratoxin A biosynthesis pathway have been identified in some producer species. However, there are few studies that purpose to identify these genes in A. carbonarius. The use of insertion mutants to identify genes associated with certain properties has been increased in the literature. Ao lado dos projetos de seqüenciamento e das análises do cariótipo pelas técnicas de hibridização in situ, o desenvolvimento de mapas genéticos fundamentados em marcadores de DNA tem propiciado consideráveis avanços à genômica de plantas. Esta revisão aborda as premissas básicas utilizadas para o mapeamento genético e suas principais aplicações, especialmente para o melhoramento vegetal. Fundamentos teóricos sobre segregação, recombinação e ligação são considerados e relacionados à construção de mapas genéticos com marcas moleculares. Abstract Microsatellites or Single Sequence Repeats (SSRs) are extensively employed in plant genetics studies, using both low and high throughput genotyping approaches. Motivated by the importance of these sequences over the last decades this review aims to address some theoretical aspects of SSRs, including definition, characterization and biological function. The methodologies for the development of SSR loci, genotyping and their applications as molecular markers are also reviewed. Finally, two data surveys are presented. Plant regeneration from proroplasts of alfalfa (Medicago sativa) via somatic embryogenesis Scientia Agricola

Alfalfa is one of the most frequently studied species from the production of tissue culture-derived embryos point of view. In this study, five alfalfa cultivars were analyzed with reference to their ability to regenerate plants from protoplast cultures via somatic embryogenesis. Plant regeneration from leaf-derived protoplasts isolated from the cultivar Rangelander was achieved using a protocol defined for alfalfa cell suspension-derived embryogenesis. Because of its high efficiency, this procedure is recommended for protoplast electroporation-mediated genetic transformation of alfalfa.

Genetic transformation is a powerful tool for plant breeding and genetical, physiological or biochemical research, consequently it is an extremely dynamic field. Transgenic plants are commonly used to complete or substitute mutants in basic research, helping the studies of complex biological situations such as pathogenesis process, genome organization, light reception and signal transduction. In this review, recent approaches for foreign gene introduction (e.g. Agrobiolistics, whole tissue electroporation, in planta Agrobacterium transformation), screening (reporter gene possibilities and performance) and transformant selection (ipt selective marker) are discussed. Reciprocal recurrent selection effects on the genetic structure of tropical maize populations assessed at microsatellite loci Genet. Mol. Biol.

A modified reciprocal recurrent selection (RRS) method, which employed one cycle of high-intensity selection, was applied to two tropical maize (Zea mays L.) populations, BR-105 and BR-106, originating the improved synthetics IG-3 and IG-4, respectively. In the present study the effects of this kind of selection on the genetic structure of these populations and their synthetics were investigated at 30 microsatellite (SSR) loci. A total of 125 alleles were revealed. A reduction in the number of alleles was observed after selection, as well as changes in allele frequencies. Rescue of a non-viable accession and rapd analysis of recovered plants of Arachis retusa PAB

In vitro regeneration of Arachis retusa was examined for the purpose of germplasm renewal and conservation. Random amplified polymorphic DNA (RAPD) fingerprinting was used to evaluate the genetic stability of plants derived from embryo axes and apical segments. Ten arbitrary decamer primers were screened and five of them were selected. Ninety genomic regions were evaluated, with an average of 18 loci per clone. All amplified segments were monomorphic. The results indicate that recovered plants are genetically stable at the assessed genomic regions and that both regeneration processes are suitable for in vitro germplasm preservation of Arachis species.
